P071 Evaluation of CFTR modulator efficacy by rectal organoid morphology analysis (ROMA) indexes

نویسندگان

چکیده

Objectives: In patient-derived rectal organoids (PDRO), CFTR modulator efficacy can be evaluated in the forskolin-induced swelling (FIS) assay. this assay, PDRO response to acute stimulation with forskolin (Fsk) and potentiator(s) is quantified as a measure of restoration function after incubation corrector(s). However, high residual and/or (eg. potentiator effect elexacaftor), may swell already during step, before any FIS measurements. The magnitude underestimated these cases. Comparison non-CF therefore also not informative. Rectal organoid morphology analysis (ROMA) indexes have been shown previously (Cuyx Thorax 2021) discriminate between from subjects without CF. Methods: were cultured for 71 ranging CF function. incubated different combinations tezacaftor, elexacaftor, ivacaftor, Fsk; acutely stimulated Fsk VX-770. ROMA calculated all conditions time points, evaluate shift range towards over time. Results: reflect changes according genotype treatment. Non-CF regardless treatment, do non-rescuable range. F508del homozygous intermediate teza-/ivacaftor, elexa-/teza-/ivacaftor. capture which no measurement performed Conclusion: provide additional information complementing example response, allowing expansion dynamic testing PDRO.

CFTR is a modulator of airway inflammation.

ALTHOUGH IT HAS BEEN 17 YEARS since the identification of the cystic fibrosis transmembrane ion receptor (CFTR) gene and protein, it remains enigmatic how abnormalities in CFTR can cause chronic and persistent pulmonary infection and inflammation that lead to bronchiectasis and end-stage lung disease (9). Because CFTR regulates transepithelial chloride and sodium conductance, the most prominent...

Optimizing Nasal Potential Difference Analysis for CFTR Modulator Development: Assessment of Ivacaftor in CF Subjects with the G551D-CFTR Mutation

Nasal potential difference (NPD) is used as a biomarker of the cystic fibrosis transmembrane conductance regulator (CFTR) and epithelial sodium channel (ENaC) activity. We evaluated methods to detect changes in chloride and sodium transport by NPD based on a secondary analysis of a Phase II CFTR-modulator study. Thirty-nine subjects with CF who also had the G551D-CFTR mutation were randomized t...

Efficacy of Rectal Misoprostol for Prevention of Postpartum Hemorrhage

Postpartum hemorrhage is an important cause of maternal morbidity and mortality after delivery. Active management of postpartum hemorrhage by an uterotonic drug decreases the rate of postpartum hemorrhage. The aim of this study is to evaluate the efficacy of rectal misoprostol for prevention of postpartum hemorrhage. This double blind randomized clinical trial was performed on full term pregnan...
